This precast polyacrylamide gel is designed for use with Criterion and Criterion™ Dodeca™ Electrophoresis Cells. It offers a versatile solution for denaturing or nondenaturing PAGE applications. The gel is 3–8% concentration, providing flexibility for different protein separation needs.
Description:
The Criterion XT Tris-Acetate Gel is made without SDS, allowing the sample buffer and running buffer to determine the separation mechanism. This makes the gel ideal for use in both native and denaturing PAGE:
- For native PAGE: Use Native Sample and Tris/Glycine Native Running Buffers.
- For molecular weight separation: Use XT Sample and XT Tricine Running Buffers.
This gel system operates at a neutral pH (pH 7.0), which helps delay acrylamide hydrolysis, offering a more stable alternative to traditional Laemmli systems.

- Ideal for molecular weight and native protein separations
Electrophoresis Mechanisms:
- Under denaturing conditions with SDS-containing Tricine running buffer, discontinuous acetate and Tricine ion fronts form moving boundaries that stack and then separate proteins by molecular weight.
- Under native electrophoresis, where proteins are separated by mass-to-charge ratio, the discontinuous acetate and glycine ion fronts create moving boundaries that stack and separate proteins by both size and charge.
